Design Notes

At the end of the riboswitch sequence (just before the fluorescent protein) we might have identified the beginning of a translated sequence coding for the CbiB protein in the natural organism. We had to insert the fluorescent protein in frame with this truncated protein coding sequence and suppress the START codon of the fluorescent protein in order to obtain a fusion fluo protein.

We got inspired by the following paper "A riboswitch sensor to determine vitamin B12 in fermented foods" Xuan Zhu 2015. The riboswitch sequence is coming from P. freudenreichii spp. shermanii DSM 20270
